The Swine Formulation of Bio-2000 was added to the primary lagoon of a 4,000 sow farrowing unit at the recommended application rate. Following the lagoon inoculation, a maintenance dose was added to each storage unit of the pull-plug system each month according to recommended application rates. Samples of the lagoon were taken before treatment and 28, 56, 133, 161 and 203 days after the initial inoculation. The sample locations and procedures are described in the following sections.
Sample Locations.
At each sampling day, vertical cross-sectional samples of the entire depth of the lagoon were collected from five locations in the primary lagoon. The five sites were located in the lagoon by east-west and north-south landmarks on the shore and were equally spaced over the entire surface of the lagoon.
Sampling Procedures.
Samples were collected from the lagoon using a 20 foot-long, hollow PVC tube (1.25 inches in diameter) fitted with a stopper at one end. The stopper was connected to a handle at the other end of the sampling tube via a metal rod running the length of the sampling tube. The sampling tube, with the stopper in the open position, was slowly lowered vertically down into the lagoon until the tube reached the bottom. At the bottom, the stopper was pulled in to the tube via the handle which trapped the column of solids and liquid in the sampling tube. The sampling tube was brought to shore and emptied by slowly releasing the stopper from the bottom of the tube. The total column of solids and liquid was collected in three 1 liter plastic containers. The solids and liquid volume in each container approximated a different 5 foot vertical column of the lagoon depth (10-15 ft., 5-10 ft and 0-5 ft). All samples were labeled by location and depth and stored on ice until chemical and micro biological analyses.
Samples were taken before treatment and at the times indicated after treatment and shipped to an independent laboratory for analysis of total solids and ammonia. Duplicate samples were analyzed for volatile fatty acids, pH and micro biologically tested for the total aerobic and facultative microorganisms as well as the specific inoculated bacteria at BioSafe Laboratories research and development laboratory. The results shown in the following table are the mean values of the five locations at the specified depth.
